Wild Type Strains

Right now we've got:

  • natural isolate

It would be nice to standardize

  • Suggestion: natural isolate from [place], [year], [PI]
  • What is the difference between 'wild type' and 'natural isolate'? Seems like strains like NC4 and AX2 should be wild types; others natural isolates. (but NC4 and AX2 and the like would possibly have PI's lab strain-- such that we would not use wild type


Laboratory Strains

Agreed on in April 2007. Summaries should include minimum information such as 'axenic'. Extra information, such as 'original lab strain' or 'sent to Loomis by Kessin' is informative and should be included.

  • AX2: Jeff Wiliams lab strain; axenic
  • AX3: Bill Loomis lab strain; axenic
  • KAX3:Jeff Hadwiger lab strain; axenic; clone of AX3 sent to Firtel by Kessin; reported to have higher transformation efficiency than AX3
  • AX4: Leandro Sastre lab strain; axenic; clone of AX3 sent to Loomis by Kessin; reported to have higher transformation efficiency than AX3
  • AX4: Adam Kuspa lab strain; axenic; clone of AX3 sent to Loomis by Kessin; reported to have higher transformation efficiency than AX3; strain that was sequenced
  • DH1: Peter Devreotes original lab strain; axenic; uracil auxotroph
  • DH1: Doug Robinson lab strain; axenic; uracil auxotroph
  • JH10: Graham Cote lab strain; axenic; thymidine auxotroph


Inviable Strains

The Summary should include the attempted mutagenesis method.

  • gene X could not be disrupted by homologous recombination
  • gene X could not be knocked down by RNAi or antisense expression
  • gene X could not be overexpressed

Reporter and Marked Strains

  • [ecmAO]:lacZ Summary: [parent strain] cells expressing lacZ under the control of the prestalk ecmAO promoter
  • [act15]:GFP Summary: [parent strain] cells expressing GFP under the control of the constitutive act15 promoter
  • Those get linked to the gene they are regulated by.


Reporter Fusions

  • [act6]:GFP:vacA Summary: GFP-vacA under the control of the act6 promoter
  • [act15]:GFP:krsA: Summary: krsA fused to GFP at the N-terminus under the control of the act6 promoter

krsA with an N-terminal GFP marker
overexpression of krsA with an N-terminal GFP marker
krsA with an N-terminal GFP marker under the control of the act15 promoter


Null Mutants

  • abcD-: Summary : abcD null mutant (no hyphen between null and mutant)
  • Add drug resistance, prototrophy or parental strain characteristics only if there are multiple strains:
    abcD-: abcD null mutant in AX3
    abcD-: abcD null mutant in JH10


REMI Mutants

  • abcD-: Summary: abcD REMI mutant
  • Jakob also has '(REMI recapitulant)' (see HS173) and '(original REMI mutant)' (see HM338) --- are those okay?


Multiple Null Mutants

  • abcD-/abcE-: Summary: abcD/abcE double null mutant
  • abcD-/abcE-/abcF-: Summary: abcD/abcE/abcF triple null mutant


Control Strains

  • [strain descriptor? Systematic Name?]: control strain for abcD- null mutant

(HR integration NOT in the target locus)

  • DO NOT link to gene


Overexpression Mutants

  • [act15]:acbD: Summary: overexpressor of abcD under the act15 promoter (and NOT 'overexpression or overexpresses')
  • abcD-/[act15]:acbD: Summary: overexpressor of abcD under the control of the act15 promoter in abcD null mutant
  • alxA-/[act15]:alxA:myc2: Summary: alxA with a C-terminal fusion to a double myc tag expressed under the control of the actin 15 promoter in an alxA null mutant; thymidine prototroph; neomycin resistant (remove last bit?)
  • Note: if you are unsure as to whether an exogenous promoter really causes overexpression of the gene, omit 'overexpressor':
    abcD-/[act15]:acbD: Summary: abcD under the control of the act15 promoter in abcD null mutant


Dominant Negative and Constitutively Active Mutants

  • Strain Descriptor: [act15]:racF2(N17T)
  • Synonym: racF2-DN
  • Summary: overexpressor of a dominant negative (hyphen?) racF2 mutation under the control of the act15 promoter

Strains with Unknown Mutation (s)

Some mutants have been isolated based on a specific phenotype but the genes responsible for the phenotype have not been characterized. The Strain Description describes that phenotype:

  • agg - (note: we currently have agg-minus; that sounds like agg minus minus)
  • slugger mutant
  • stalky mutant
  • tester strain (what does that mean?)


Others

  • "pkgB null expressing pkgB(K182M) (predicted kinase dead mutant) under the control of the act15 promoter" (pkgB-/[act15]:pkgB(K182M) strain link)
